Rolling Circle Amplification to clone circular viral genome
Rolling circle amplification (RCA) is an isothermal enzymatic process in which a random
primer is used to amplify a circular DNA template by a special DNA polymerase called phi29
DNA polymerase. In recent years, this technique has been used for the efficient amplification
of circular DNA viral genomes without the need of specific primers. The viral genomes can
be further cloned for characterisation and infectious genomic clones can be generated directly
from the samples. This will help in the detection of novel viruses. During the workshop, the
participants will be learning the techniques involved in the process of cloning circular DNA
